Опухолевые поражения костей – гетерогенная патология, которой свойственно агрессивное</p><p>течение, частые рецидивы и низкие показатели выживаемости. Разработка и внедрение эффективных и новаторских методов лечения остаётся актуальной задачей. Радикальная интраоперационная термоабляция (РИТ), основанная на высокотемпературном воздействии, представляет собой новый оригинальный метод лечения. Настоящее исследование направлено на изучение влияния термоабляции (ТА) на клеточную гибель.</p><p>Цель исследования – оценить влияние ТА (с использованием метода РИТ) на жизнеспособность опухолевых клеток.</p></sec><sec><title>Материал и методы</title><p>Материал и методы. В исследование было включено 8 пациентов с опухолевым поражением костей скелета. Опухоли подвергались ТА при температуре 60 °C в течение 30 мин ex vivo. Исследовались образцы опухолевой ткани до и после сеанса термоабляции. Для оценки апоптоза использовались два метода: проточная цитометрия и метод TUNEL.</p></sec><sec><title>Результаты</title><p>Результаты. Радикальная интраоперационная термоабляция, разработанная нами, является многообещающим методом лечения опухолевого поражения костей. Температурный режим 60 °C в течение 30 мин эффективно инициирует процессы гибели клеток в опухолевой ткани, что подтверждается значительным увеличением числа апоптотических клеток сразу после сеанса термоабляции и достоверным ростом количества клеток с признаками позднего апоптоза через час после ТА. Применение метода TUNEL подтверждают результаты проточной цитометрии, демонстрируя существенное увеличение числа клеток на поздних стадиях апоптоза. Полученные нами данные свидетельствуют о выраженном противоопухолевом эффекте сразу после проведения РИТ.</p></sec><sec><title>Заключение</title><p>Заключение. Результаты исследования подтверждают, что РИТ представляет собой перспективный органосохраняющий метод лечения опухолевого поражения костей, требующий дальнейшего изучения для расширения его применения в клинической практике.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Introduction</title><p>Introduction. Bone tumors are heterogeneous group of skeletal neoplasms characterized by frequent recurrences, aggressive clinical course and low survival rates. The development of new treatment methods continues to pose pressing challenges. Radical intraoperative thermal ablation (RIT) using high-temperature exposure is emerging as a new and promising strategy for organ-preserving treatment. This study focuses on the effect of thermal ablation (TA) on tumors.</p><p>Objective of the Study to assess the impact of TA (using the RIT method) on the viability of tumor cells.</p></sec><sec><title>Material and Methods</title><p>Material and Methods. The study included 8 patients with bone tumors. Tumors underwent TA at a temperature of 60 °C for 30 minutes ex vivo. Apoptosis was studied in tumor tissue samples before and after TA. Apoptosis was assessed using two methods: flow cytometry and the TUNEL assay.</p></sec><sec><title>Results</title><p>Results. The TA procedure developed by our scientific group represents a promising organ-preserving approach for treating malignant bone tumors. A temperature regime of 60 °C for 30 minutes was effective in initiating tumor cells death. This was confirmed by two independent methods – flow cytometry and TUNEL assay – which demonstrated a significant increase in the number of apoptotic cells immediately following the procedure and a notable rise in the number of cells exhibiting signs of late apoptosis one hour post thermal ablation. Therefore, the collected data confirm a pronounced antitumor effect immediately after implementing RIT.</p></sec><sec><title>Conclusion</title><p>Conclusion.
